Quarterback Charlie Batch led Pittsburgh (3-1), throwing for 102 yards, including a 37-yard first-quarter touchdown to Emmanuel Sanders.
Panthers' third-string quarterback Jimmy Clausen also had an impressive night, throwing for 173 yards and two scores.
Quarterback Cam Newton, wide receiver Steve Smith and many of the Panthers' first-string starters did not play in the contest, as Carolina (2-2) awaits its regular-season opener next Sunday at Tampa Bay.
Carolina linebacker Jon Beason was also held out, as he continues to deal with a hamstring injury suffered in training camp.
Clausen's 79-yard touchdown pass to Lamont Bryant poised the Panthers to tie the Steelers late in the fourth quarter, but coach Ron Rivera chose to go for the two-point conversion.
The attempt failed and just over two minutes later, Medlock's long field goal attempt missed to give Pittsburgh the win.
